The first missionary called was in 1841, but did not fulfill his mission. In 1851 Parley P. Pratt, Rufus C. Allen, and Philo Bennett Wood were called to serve in Chile, becoming the first missionaries in South America. Their mission was largely unsuccessful. 

These individuals were not part of a official mission, instead serving in various locations for varied amounts of time.

The first official mission in South America was established in 1925 with the headquarters in Buenos Aires after German immigrant families who had joined the Church in Europe settled in Argentina.
